      目的以正常人肝L-02细胞建立的脂肪变性肝细胞模型为研究对象,观察姜黄素对肝细胞胆固醇合成及转运的影响。方法用MTT法观察姜黄素对脂肪变性人肝L-02细胞模型增殖的影响,用油红O染色定性观察细胞内脂滴形成情况;逆转录-多聚酶链反应(RT-PCR)半定量检测胆固醇合成限速酶HMG-CoA还原酶及转运蛋白Caveolin-1在mRNA水平的表达;Western-blotting法检测细胞caveolin-1的表达;高效液相色谱法(HPLC)定量检测细胞内外胆固醇各组分含量。结果姜黄素呈浓度和时间依赖性减少细胞内脂滴数量,降低脂变肝细胞内总胆固醇(TC)、胆固醇酯(CE)含量,增加培养基中游离胆固醇(FC)含量,同时减少HMG-CoA还原酶的表达,增加Caveolin-1 mRNA的表达,并且呈浓度时间依赖性增加caveolin-1的蛋白表达。结论姜黄素降低胆固醇作用可能与Caveolin-1的上调、HMG-CoA还原酶的下调有关。
